Averting inflammation by targeting the cytokine environment

In 1957, a soluble factor that protected cells from viral infection was discovered by Isaacs and Lindenmann and named interferon (IFN)1, 2. This landmark finding set the stage for cytokine research. Since then more than 90 cytokines and cytokine receptors have been identified, nine of which are the basis for current therapeutics on the market3. Given the fundamental roles of cytokines in the development and pathogenesis of many inflammatory diseases, there has been extensive worldwide research and development focused on blocking or enhancing cytokine activity.

There are numerous clinical applications for targeting cytokines and include inflammatory diseases, cancer immunotherapy, bone disorders, metabolic diseases, wound healing and antiviral therapy. Although not all potential cytokine targets can be covered in this Review, we will outline some of the most effective and promising cytokine targets that have been linked to inflammatory diseases in preclinical and clinical studies in recent years. This selection is based on an established overview of the literature and is shaped by a viewpoint of the probable success or failure of targeting these cytokine pathways. Finally, we will discuss some of the issues facing the development of cytokine-targeted drugs with respect to the type of therapeutics being developed and the inherent redundancies in inflammatory cytokine networks.

Berries – the secret brain food?

Linda’s comments: We in the holistic/alternative world have known this for years….what this article has forgotten to bring to your attention is, YOU MUST EAT ORGANICALLY.  Doesn’t do you any good eating fresh fruits that are laden with pesticides and herbicides and who knows what else. The U.S. Department of Agriculture   wants to pat themselves on the back, but do you see where they WARN you about the GMO, pesticides/herbicides??  Hmmmmm….they can’t be this brainless can they?  On the other hand could it be they don’t care and only care about their bottom-lines? They want to talk about “leading to brain disorders, heart disease and cancer, among other degenerative conditions” FAILING to tell you that you are in more danger eating GMO, pesticides/herbicides.

Yes, I ENCOURAGE all of you to eat healthy and NON-GMO.  AND, YES, I will continue encourage you to eat organically without nasty pesticides And herbicides.

People who are closely related tend to have similar bacteria in their intestines. But a study of the viruses in people’s faeces has found that even identical twins have uniquely different populations of these tiny tenants.

Florent Angly, a biologist at the University of Queensland and a co-author of the study, said the findings showed that factors such as health and diet were the biggest influence on the kinds of viruses people harboured.

”Twins have an identical genome, so including twins in the study was designed to find out what is more important: nature or nurture,” he said.

Humoral Immune Response in Dogs with Borrelia

Lyme arthritis in dogs can be induced under experimental and
natural conditions.
However, the veterinary relevance of canine borreliosis is still
under extensive investigation. The prevalence of symptoms is
clearly low although the risk of tick exposure is high. Current
research focuses on case definitions, methods for diagnosing
clinical disease in dogs, and discrimination between an immune
response to a natural infection versus vaccination.
In this experimental study,
23 dogs raised under tick-free conditions were allocated to two
groups: 11 dogs were vaccinated with a commercial borrelia
vaccine and subsequently developed detectable antibody titers; 12
were walked in a tick-endemic area on two consecutive days. On
day five after exposure engorged ticks were removed from the 12
dogs and analyzed for Borrelia DNA in real-time PCR assay. Blood
samples were taken before exposure/vaccination and at defined
time points thereafter.

Mercury-induced inflammation

Methods
Human leukemic cultured LAD2 mast cells and normal human umbilical cord blood-derived cultured mast cells (hCBMCs) were stimulated by HgCl2 (0.1-10 microM) for either 10 min for beta-hexosaminidase release or 24 h for measuring vascular endothelial growth factor (VEGF) and IL-6 release by ELISA.
Results
HgCl2 induced a 2-fold increase in beta-hexosaminidase release, and also significant VEGF release at 0.1 and 1 microM (311+/-32 pg/10*6 cells and 443+/-143 pg/10*6 cells, respectively) from LAD2 mast cells compared to control cells (227+/-17 pg/10*6 cells, n=5, p<0.05). Addition of HgCl2 (0.1 microM) to the proinflammatory neuropeptide substance P (SP, 0.1 microM) had synergestic action in inducing VEGF from LAD2 mast cells. HgCl2 also stimulated significant VEGF release (360 +/- 100 pg/10*6 cells at 1 microM, n=5, p<0.05) from hCBMCs compared to control cells (182 +/-57 pg/10*6 cells), and IL-6 release (466+/-57 pg/10*6 cells at 0.1 microM) compared to untreated cells (13+/-25 pg/10*6 cells, n=5, p<0.05). Addition of HgCl2 (0.1 microM) to SP (5 microM) further increased IL-6 release.
Conclusions
HgCl2 stimulates VEGF and IL-6 release from human mast cells. This phenomenon could disrupt the blood-brain-barrier and permit brain inflammation. As a result, the findings of the present study provide a biological mechanism for how low levels of mercury may contribute to ASD pathogenesis.

Association of Carotid Plaque Lp-PLA2 with Macrophages & Infection

The significant association of plaque Lp-PLA2 with plaque macrophages and C. pneumoniae suggests an interactive role in accelerating inflammation in atherosclerosis. A possible mechanism for C. pneumoniaein the atherogenic process may involve infection of macrophages that induce Lp-PLA2 production leading to upregulation of inflammatory mediators in plaque tissue. Additional in vitro and in vivo research will be needed to advance our understanding of specific C. pneumoniae and Lp-PLA2 interactions in atherosclerosis.

Deadly inflammation, but no sign of infection

Injury causes a systemic inflammatory response syndrome (SIRS) that is clinically much like sepsis. Microbial pathogen-associated molecular patterns (PAMPs) activate innate immunocytes through pattern recognition receptors. Similarly, cellular injury can release endogenous ‘damage’-associated molecular patterns (DAMPs) that activate innate immunity. Mitochondria are evolutionary endosymbionts that were derived from bacteria and so might bear bacterial molecular motifs. Here we show that injury releases mitochondrial DAMPs (MTDs) into the circulation with functionally important immune consequences. MTDs include formyl peptides and mitochondrial DNA. These activate human polymorphonuclear neutrophils (PMNs) through formyl peptide receptor-1 and Toll-like receptor (TLR) 9, respectively. MTDs promote PMN Ca(2+) flux and phosphorylation of mitogen-activated protein (MAP) kinases, thus leading to PMN migration and degranulation in vitro and in vivo. Circulating MTDs can elicit neutrophil-mediated organ injury. Cellular disruption by trauma releases mitochondrial DAMPs with evolutionarily conserved similarities to bacterial PAMPs into the circulation. These signal through innate elicit neutrophil-mediated organ injury identical to those activated in sepsis to create a sepsis-like state. The release of such mitochondrial ‘enemies within’ by cellular injury is a key link between trauma, inflammation and SIRS.

Copper research

Copper is essential for many functions. This is a new one; healthier prions with less risk of prion diseases. It seems that it stabilizes Prions. This means they are less likely to misfold or aggregate when copper is present.

Low levels of copper have been tied to arrhythmias and many other health problems. It is best to NOT restrict copper unless there is proven copper excess, and serum copper is merely a reflection of inflammation and does not reveal elevated copper levels in the tissues.
